This recipe was devised as an accompaniment to?spatchcock stuffed chicken. By cooking the beets over a long period of time with a low amount of heat, the natural sugars in the vegetable caramelize producing a sweetness which is echoed by the balsamic reduction. This dish almost didn?t make it to the table due to my spring-loaded oven doors. Luckily a quick catch saved the beets which proved to be a perfect pairing with the poultry.

Slow-Roasted Beets

This dish would be excellent with a handful of nuts thrown in at the end (walnuts or almonds) and feta could be substituted for the goat cheese.

Yield: 6 servings

4 large beets
1 onion, sliced
1 tablespoon olive oil
pinch of sea salt and ground pepper
1/2 teaspoon dried oregano
1/2 teaspoon dried thyme
1/2 teaspoon dried chili flakes
4 tablespoons balsamic vinegar, divided
2 tablespoons butter
1/3 cup crumbled goat cheese
small handful of fresh parsley, torn

Preheat oven to 350 degrees F. (180 degrees C.)

In a large roasting pan, add the beets, onion, olive oil, salt, ground pepper, dried oregano, thyme, chili flakes and balsamic vinegar. Toss to coat and disperse ingredients. Spread out in a single layer at the bottom of the pan and dot the top with butter.

Roast for 1 hour. Drain off liquid into a small sauce pan and reserve. Return to oven for an additional 2-1/2 hours. Stir occasionally.

Over a medium heat, add additional balsamic vinegar and simmer until thickened, around 3 minutes. Swirl pain occasionally to keep from burning.

Drizzle over balsamic reduction. Add goat cheese and parsley and gently toss. Serve warm or room temperature.

Sears refurbishes four stores with new concept in Canada

Barrie, Belleville, Newmarket and Hamilton are the First - With a renewed commitment to customer service and dramatic changes to store layout and merchandising, big things are happening at Sears.

On May 25th, Sears Canada unveils its new, refreshed store concept in four Ontario locations: Barrie, Belleville, Newmarket and at the Lime Ridge mall in Hamilton. These stores reflect the Company's plans to transform the Sears shopping experience in Canada.

Changes in the stores include: wider aisles, less clutter and streamlined merchandise offerings with exciting new brands at everyday low prices. In all the stores, expanded sections include: KidsRoom and BabyRoom, Jessica women's suit shops, Men's suit shops with easier by-size shopping and additional fitting services, a new assortment and look for Craftsman outdoor power equipment and a market-leading assortment of major appliances and mattresses.

In Newmarket, the new store has an improved check-out process. In Barrie and Belleville, easy-access shopping carts will now be available.

In addition, Trend Directors Cynthia Florek (Womenswear) and Nancy Dennis (Menswear and Kidswear) will be on hand in two of the stores on the 26thto present special style-savvy seminars. Cynthia and Nancy will be in Newmarket at 11am and in Barrie at 2pm.

India PM seeks to heal bad blood on Myanmar visit

SITTWE, Myanmar (Reuters) - In northwest Myanmar, where the Kaladan River flows out into the Bay of Bengal, the two giant arms of a half-built wharf enfold the estuarine mud with steel and concrete.

Their embrace is fraternal - Myanmar's giant neighbor India is funding the new port in Sittwe, the capital of Rakhine State - but also strategic.

The port is part of a $214-million river and road network that will carve a trade route into India's landlocked northeast and underscore New Delhi's determination to capitalise on Myanmar's growing importance at Asia's crossroads.

Manmohan Singh will seek to bolster ties this week during the first visit by an Indian prime minister to Myanmar in 25 years. His official agenda includes road, rail, waterways and air links, says India's foreign ministry.

Unofficially, he must also overcome a history of bad blood with Myanmar, where Indian investments are already dwarfed by regional rival China.

The visit follows a year of dramatic reforms in which Myanmar has pulled back from China's powerful economic and political orbit and won a suspension of U.S. and European sanctions. With change has come a series of high profile visits, including stops by the leaders of Britain and South Korea, as well as U.S. Secretary of State Hillary Clinton.

President Thein Sein's government has held peace talks with ethnic minority rebels, relaxed strict media censorship, allowed trade unions and protests and held a by-election dominated by Nobel Peace Prize laureate Aung San Suu Kyi's opposition party.

As Myanmar emerges from decades of isolation, trade with its neighbour is already swelling. Myanmar's government expects two-way trade with India to nearly double in two years to $2 billion, from $1.4 billion in the year to March 30, a figure that was nearly 30 percent higher from the previous year, according to Myanmar's Ministry of Commerce.

"Stronger trade and investment links, development of border areas, improving connectivity between our two countries and building capacity and human resources are areas that I hope to focus on during my visit," Singh said in a statement released before his arrival in the capital Naypyitaw on Sunday evening.

India should be a natural partner, with ties stretching back to the ancient Buddhist emperor Ashoka and, more recently, a shared experience of British colonialism and World War Two.

But its business interests in the former Burma have been "few and far between" since the mass expulsion of Indian merchants after the military seized power in 1962, says Thant Myint-U, author of "Where China Meets India: Burma and the New Crossroads of Asia".

"Many in India remember all too well that this was the country that nationalized Indian businesses and expelled hundreds of thousands of ethnic Indians with literally nothing more than the shirts on their backs," he said.

One hopeful symbol of improved ties is Sittwe.

The two countries formally agreed on the so-called Kaladan Multimodal Transit Transport Project in April 2008, just seven months after Myanmar's military junta crushed nationwide pro-democracy protests led by Buddhist monks.

Work began on Sittwe port in September 2010, shortly before the former military junta held a rigged election that brought to power a quasi-civilian but surprisingly reformist government.

Indian conglomerate Essar Group is building the port on 70,000 square metres (753,000 sq ft) of landfill in Sittwe's centre. It should be ready in two years, says Myanmar's Commerce Ministry, accommodating ships from the Indian city of Kolkata, a 539-km (334 mile) voyage away across the Bay of Bengal, and handling up to 500,000 metric tons a year.

From Sittwe, ships will sail up the Kaladan River to the town of Paletwa, where Essar will build a second, smaller port. A 122-km (76-mile) highway will connect Paletwa to the Indian state of Mizoram. The two ports and dredge work will cost $74 million. The highway will cost $140 million.

STRATEGIC SIGNIFICANCE

India is already Myanmar's third-biggest export market after Thailand and China. But Thant Myint-U plays down Myanmar's economic importance to India. "Myanmar is extremely important for India's northeast, but because the northeast itself rarely gets Delhi's attention, that in itself doesn't count for much."

However, New Delhi is acutely aware of Myanmar's strategic significance "because of China's increasing economic presence and anxiety about a possible future Chinese presence on the Bay of Bengal", he says.

Not far south of Sittwe, Chinese money is funding a bigger port and special economic zone in Kyaukphyu, a coastal town where Myanmar-China pipelines reach the Bay of Bengal, creating a passage from western China to South and Southeast Asia and allowing shipments of fuel and natural resources to avoid the Malacca Strait.

An Essar company official said "communication problems" had been a headache during the project, with Myanmar officials slow to provide information and language issues also a hurdle. The company, however, would consider further projects in the country, given they had already worked there.

"Business is all about relationships, and we have been meeting the right people," the official said.

A delegation of Indian business officials will join the prime minister on his visit, said an Indian Foreign Ministry official, adding that India was looking at setting up a special economic zone.

Indian companies are showing interest too.

Tata Motors, which makes the ultra-cheap Nano and owns Jaguar Land Rover, is looking to expand operations in Myanmar by assembling and selling buses and light commercial vehicles there, its India head said in January. Tata already operates a truck assembly plant in Myanmar.

State-run explorer Oil and Natural Gas Corporation Ltd, which has two gas blocks in Myanmar with production due to start in 2013, and tractor maker Escorts Ltd have also said they are looking to expand operations there.

Myanmar opposition leader Aung San Suu Kyi was once lionised by New Delhi, which gave her the prestigious Jawaharlal Nehru Award in 1993 and Myanmar's dictatorship the cold shoulder. Singh will meet Suu Kyi in the biggest city Yangon on Tuesday.

But with growing investment in Myanmar by regional rival China, the world's biggest democracy has forged closer ties, inviting former dictator Senior General Than Shwe on an official state visit to India in 2004.

Three years later, after he presided over a violent crackdown on pro-democracy protests led by Buddhist monks, India was widely criticized for its muted response amid international outrage.

Skp2 activates cancer-promoting, glucose-processing Akt

ScienceDaily (May 26, 2012) ? HER2 and its epidermal growth factor receptor cousins mobilize a specialized protein to activate a major player in cancer development and sugar metabolism, scientists report in the May 25 issue of Cell.

This chain of events, the scientists found, promotes Herceptin resistance in breast cancer and activation of glucose metabolism (glycolysis), which cancer cells primarily rely on to fuel their growth and survive.

Their research focused on Skp2 E3 ligase, a protein that binds to and tags other proteins with molecules called ubiquitins, in this case to activate the Akt kinase. "We discovered a novel function of Skp2 E3 ligase that makes it an important player in cancer development and also identified a crucial role for it as a regulator of the glycolysis pathway," said senior author Hui-Kuan Lin, Ph.D., associate professor in MD Anderson's Department of Molecular and Cellular Oncology.

"This is potentially important for understanding and addressing Herceptin resistance in breast cancer," Lin said. "The effect on glucose metabolism also could have implications for other types of solid tumor cancers, including prostate, because they rely so heavily on glycolysis."

The team also found that Skp2 overexpression is associated with poor prognosis for breast cancer patients and its spread to other organs.

Lin and colleagues are studying potential inhibitors of Skp2 that might be developed for treatment.

The EGFR family of proteins includes HER2, which abundantly coats cancer cells in about a third of breast cancers, making these tumors prime targets for the targeted drug Herceptin. Fundamental discovery about Akt activation

The Akt kinase relays signals by growth factors from outside of the cell into the cell. It regulates cell profliferation and survival, metabolism and tumor development, the authors noted.

To do its work, whether normal or oncogenic signaling, it must move from the cytosol to the plasma membrane. To do that, Lin and colleagues had previously shown that Akt must be ubiquitinated -- and those ubiquitins must be attached in a specific chain formation, the K63-linked polyubiquitin chains.

That earlier finding involved the insulin-like growth factor receptor (IGF-1) and a different E3 ligase. "Finding that the epidermal growth factor receptors also ubiquitinate Akt, and that they do so through the Skp2 E3 ligase, was quite unexpected," Lin said.

Finding two paths to ubiquitination implies that there might be more, Lin said.

Regulating cancer's energy engine

Tumor cells evolve to rely mainly on glucose processing to generate energy, grow and thrive. They increase glucose uptake and glycolysis, a process that doesn't involve oxygen and occurs in the cell's cytosol. Non-cancerous cells rely more heavily on oxygen-based metabolism of fatty acids and other nutrients in the cell's mitochondria. This difference is called the Warburg Effect.

In a series of experiments, the researchers blocked Skp2 expression and found:

? Suppression of glucose uptake and glycolysis in breast cancer cell lines.

? Reduction of tumor size and glucose uptake in a mouse model of HER2-expressing breast cancer.

? Steep declines in the expression of glucose-transporter Glut1. Cells with Skp2 produced more Glut1 when induced by epidermal growth factor signaling. Akt activates Glut1.

? Reduced Akt activation, Glut1 expression and tumor growth plus extended survival in mice genetically engineered to develop breast cancer and metastasis.

Poor prognosis, Herceptin resistance

The team analyzed 213 cases of breast cancer treated with modified radical mastectomy but no chemotherapy or radiation. Of these, 80 were HER2-positive. In these tumors Skp2 expression was correlated with:

? Adverse prognostic factors such as cancer stage, primary tumor status and lymph node metastasis.

? Increased activation of Akt.

A multivariable analyses of prognostic factors showed Skp2 overexpression to be a significant factor affecting metastasis-free survival for HER2-positive breast cancer patients. No prognostic role was found for Skp2 expression among patients with HER2-negative disease. HER2-positive breast cancer patients can develop resistance to Herceptin treatment, the researchers noted, and some encounter this problem right at the start. Knocking down Skp2 made cancer cell lines more susceptible to Herceptin, stifling cancer proliferation. In another mouse model, Herceptin by itself inhibited tumor growth but did not shrink tumors. With Skp2 inhibited, Herceptin treatment caused the tumors to nearly vanish in 22 days.

This sensitivity was blocked when the scientists overexpressed Akt in Skp2-silenced cancer cells.

Overall, the team's findings indicate that blocking Skp2 might inhibit glycolysis and that targeting glycolysis could be an important approach to cancer treatment, Lin said. Syria 'massacre' raises pressure for international response

Syrian artillery attack that apparently killed more than 90 people, including many children, brought French condemnation, while the UK said it would call for urgent UN Security Council meeting.

A Syrian artillery barrage killed more than 90 people, including dozens of children, in the worst violence since the start of a UN peace plan to staunch the flow of blood from Syria's uprising, activists said on Saturday.

The bodies of children were shown in footage posted to YouTube purporting to show the victims of the shelling in the central town of Houla on Friday. The sound of wailing filled the room.

The reports of the carnage, which could not be confirmed independently, underlined how far Syria is from any negotiated path out of the 14-month-old revolt against President Bashar al-Assad.

French Foreign Minister Laurent Fabius condemned the violence as a "massacre," and said he wanted to arrange a meeting in Paris of the Friends of Syria, a group that brings together Western and Arab countries keen to remove Mr. Assad.

Syrian state television aired some of the footage disseminated by activists, calling the bodies victims of a massacre committed by "terrorist" gangs.

A British-based opposition group, the Syrian Observatory for Human Rights, said residents of Houla were fleeing in fear of more shelling.

It said one person was killed in the northern town of Saraqeb when security forces opened fire on a protest against the killing. Activists distributed footage appearing to show similar protests in Aleppo, the largest city in the north.

FAMILIES KILLED

A member of the fragmented exile group that says it speaks for Syria's political opposition said Assad's forces had killed "entire families" in Houla in addition to the shelling.

"The Syrian National Council (SNC) urges the UN Security Council to call for an emergency meeting ... and to determine the responsibility of the United Nations in the face of such mass killings," SNC spokeswoman Bassma Kodmani said.

Opposition activists said Syrian forces had opened fire with artillery on Friday after skirmishing with insurgents in Houla, a cluster of villages north of the city of Homs, itself battered by shelling.

Although Annan's six-week old ceasefire plan has failed to stop the violence, the United Nations is nearing full deployment of a 300-strong unarmed observer force meant to monitor a truce.

Morgan Stanley is prepared to refund retail Facebook IPO investors who purchased company stock last Friday, a source familiar with the matter told USA Today. The lead investment bank in Facebook?s initial public offering plans to compensate investors who reportedly overpaid.

Facebook IPO Investors Suffer Major Losses

Investors were excited about purchasing stock on Facebook IPO day, but were quickly disappointed to learn that trading was stalled by 30 minutes as a result of technical problems on the Nasdaq Stock Market.

As a result of the major glitch, investors complained that their orders were never processed correctly, which resulted in losses. Some larger market makers say their losses have?reached as high as?$100 million.

Additionally, Facebook and Morgan Stanley are facing lawsuits after the IPO fiasco. Investors alleged that analysts at the large underwriting investment banks cut their second-quarter and full-year forecasts for Facebook just before the IPO, but told?only a handful of clients.

Morgan Stanley to?Compensate Individual Investors

While Morgan Stanley has declined to comment about the lawsuits it faces,?it plans to compensate its retail investors, also known as individual investors, who paid too much on Facebook IPO day.

The source who shared the company?s plans said the firm is reviewing the orders its individual clients placed for Facebook stock, and will make price adjustments if the clients paid too much.

The person, who spoke on the condition of anonymity since they were not authorized to discuss the matter publicly, was unsure of what amount constituted overpayment. However, an investor shared that they?d placed an order at 8 a.m. on May 18 but didn?t receive confirmation of that trade until Monday (May 21) when the price was much higher than the opening bell.

Facebook stock, which fell flat on IPO day by closing?at $38.23 per share, has not experienced impressive results. On Friday morning, the stock was down 15 percent from the price of its initial public offering a week prior, trading at just $32.57 per share.

Make a real effort to stop worrying. Worrying unnecessarily will only create worst-case scenarios in your mind. Instead of worrying, think about the worst thing that could happen, and prevent it. Then, you are prepared to handle whatever gets thrown your way. Excessive worrying solves nothing.

Never force yourself to work beyond your physical limitations as you pursue your goals. Doing your best is essential, but you also need to know what your best is and not go past it. Caring for yourself is important when you are striving towards a desired end. If you were to sacrifice your body in pursuit of improving yourself, it would be counter-productive to your needs.

Personal development begins when you identify what you value the most. Setting high standards for yourself can help you achieve personal development you desire.

When building a personal development plan, it is important to consider your personal values. It's counterproductive to focus on things that you don't value. Instead, work on aspects of your life you want to improve that mesh well with your individual beliefs. This will help you make significant changes to your life, both at home and at work.
